Abstract:

Background: Chronic ethanol abuse causes reproductive organ failure and infertility in both humans and laboratory animals. Since sperm has a critical role in reproductive function, the objective of this unique study was to evaluate the effects of different doses of ethanol on sperm parameters, chromatin structure and apoptosis in adult mice. Materials and Methods: A total of 36 adult male mice were equally divided into four groups. Group 1 received ethanol (10%, v/v) containing saccharin (0.2%, w/v), group 2 received ethanol (5%, v/v) containing saccharin (0.1%, w/v), group 3 was treated with saccharin (0.2%, w/v) and group 4 served as control and fed on basal diet for 35 days. Finally, the left cauda epididymis of each animal was cut and placed in Ham’s F10 medium. Retrieved spermatozoa were used to analyze count, motility, morphology and viability. Sperm chromatin condensation and DNA integrity were assessed by five different tests including chromomycin A3 (CMA3), toluidine blue (TB), sodium dodecyl sulfate (SDS), and SCD (sperm chromatin dispersion), and sperm apoptosis was assessed by TUNEL. Results: Following ethanol consumption, the sperm count diminished in the ethanol-treated groups. A decrease in sperm motility and an increase in the rate of morphological abnormalities (coiled and broken tails) were seen in the experimental and saccharin groups in comparison with controls. We showed that ethanol consumption can disturb sperm DNA integrity and chromatin remodeling and it may also induce sperm apoptosis. The rates of sperm apoptosis were 51.57 ± 7.45 and 42.85 ± 6.76 in the high ethanol dose and low ethanol dose groups, respectively. Conclusion: The results showed that alcohol has negative effects on sperm parameters, chromatin/DNA integrity and apoptosis in mice. These alcohol-induced sperm anomalies may be dose-dependent.

P-76: Effects of Concurrent Ethanol and Saccharin Consumption on Sperm Parameters in Adult Mice

Background: Since sperm parameters have a critical role in reproductive function, the objective of this study was to evaluate the effects of concomitant ethanol and saccharin consumption on sperm parameters in adult mice. Chronic ethanol consumption suppresses male reproductive function and sexual behavior in laboratory animals and humans. Furthermore, alcohol abuse by males has been correlated...

P-11: The Effects of Kerack Used in Iran on Sperm Parameters and Testis Structure in Adult Mice

Background: Kerack is emerging illicit substance which its use is rising up in Iran. It has harmful effects on body organs. The aim of this study is to investigate the effects of Iranian Kerack on sperm parameters and testicular structure of mice. Materials and Methods: In this study, 25 male mice (Balb/C) were divided into five groups (control, sham and three experimental). Experimental groups...

P-54: Evaluation of The Acute and Chronic Effects of Different Gold Nanoparticle Doses on Sperm Parameters and Chromatin Structure in Mice

Background The solid and colloidal particles in the range of 1-100 nm are called nanoparticles. Recently, one of the most important metal nanoparticles, with wide usage is gold nanoparticle. Based on the previous studies, gold nanoparticles have spermatotoxic effects and can destroy sperm motility. The aim of present study is the effect of gold nanoparticles on chromatin quality and sperm param...

P-73: The Effects of Manganese on Testis Structure and Sperm Parameters in Adult Mice Exposed to Formaldehyde

Background: Formaldehyde (FA) as a ubiquitous environmental pollutant is extensively used in hospitals, laboratories and many industrial settings. FA exerts adverse effects on testicular structure and sperm param eters through increasing oxidative stress. Manganese, a well-known antioxidant, can inhibit oxidative stress damages. The aim of this study was to investigate the influences manganese ...

P-61: Effects of Acrylamid on Sperm Parameters and Chromatin Quality in Mice

Background: Acrylamid (AA) is found in carbohydraterich foods that are prepared at high temperatures, such as French fries and potato chips. The toxic effects of AA in male animals include degeneration of epithelial cells of seminiferous tubules, decreased number of sperm and elevation in abnormal spermatozoa. The aim of this study was to evaluate the detrimental effects of AA on sperm paramete...
